Remove Trojan.Autoit.CKU Virus

Hello, this is urgent! Can you help me remove Trojan.Autoit.CKU virus from my work computer? I have tried the antivirus, but it didn’t help me anything. The virus always comes back every time when I restart computer after I ran antivirus to kill it. I cannot finish my work on computer now. When i logged on a pop up occurred with an audio message that stated that i was in risk with Trojan.Autoit.CKU infected and that Microsoft had locked my computer so no other damage could be done. It instructed to call (888) 343 – XXX with. Is this a legitimate number to Microsoft as i have F-Secure on my PC? I am afraid some folders about my work information would be corrupted by this virus, so please help me remove this virus now.
